Zinc transporter 1 (ZnT1) and ZnT4 are two high-affinity zinc transporters expressed in pancreatic beta cells. ZnT1 and ZnT4 share 90% homology in their N-terminal transmembrane domains but differ in their C-terminal regions. To investigate the potential role of ATP in the expression of ZnT1 and ZnT4, insulinoma cells were treated with low glucose, high glucose, and KCl, and the expression of ZnT1 and ZnT4 were measured. Low glucose and high glucose markedly increased ZnT1 and ZnT4 expression at mRNA and protein levels. ATP depletion reduced glucose-induced increases in ZnT1 and ZnT4 at both mRNA and protein levels. ATP injection increased the expression of ZnT1 and ZnT4 in low glucose. Excess KCl did not influence the expression of ZnT1 or ZnT4. Insulin increased ZnT1 and ZnT4 expression, which could be attenuated by ATP depletion. The present data demonstrated that ATP depletion could reduce the increase of ZnT1 and ZnT4 induced by high glucose. These results indicate that ATP may play a role in regulating the expression of ZnT1 and ZnT4 in pancreatic beta cells.Q:
